Lab 1A: DNA Isolation from Wheat Germ Sherry Hebert BIOL-3221L-U2020F Lab Procedure Overview ● Homogenization of wheat germ:

physical disruption of tissues/cells ● Removal of lipids and dissociation of DNP complexes ● Removal of proteins ● Precipitation of DNA Homogenization 5 g wheat germ is blended with 30 mL of warm DNA buffer Centrifugation 10 min @ 4000 x g Discard supernatant Lipid Removal Resuspend in 5 mL DNA buffer + 4 drops Triton-X This solubilizes membranes/lipids & results in dissociation of the DNP complexes (separates DNA & associated proteins) Centrifugation 15 min @ 4000 x g Discard supernatant DNA and proteinlipids Protein Removal Resuspend in 30 mL 2.6M NaCl This allows the precipitation of proteins, while the DNA remains in solution Centrifugation 15 min @ 20,000 x g KEEP supernatant proteinDNA Isolate DNA Carefully add 30 mL 70% ethanol This allows the precipitation of DNA at the interface interfaceEtOH salt solution Collect Collect DNA by rapidly stirring and place on plastic Petri dish Clean Remove liquid, wash with 10 mL EtOH, and again remove liquid.
