Agar seems to be covering about Half (~50%) of the Surface of its Bottom

Agar Plate. What's going on?

Buffy's Lab Notebook. You ask this Clan a Series of Questions:

• "How much Top Agar did you add to each of the small Test Tubes in the

Heating Block?" Alfonso says approximately 1.5 ml.

• "How much of each Bacteriophage Dilution did you add to each of the small

Test Tubes when you removed them from the Heating Block?" Buffy

answers that she added exactly 100 μl.

• "How much E. coli did you add to each of the small Test Tubes after the

Bacteriophage Dilution was added?" Bucky says he added exactly 200 μl.

• "Did you vortex each of the small Test Tubes after the Bacteriophage and the

E. coli were added and did you pour all 1.5 ml of Top Agar into the Bottom

Agar Plate?" Alfonso answers Yes.

• "How did you create these Half Top Agars?" Alfonso says he poured the

Contents of each Top Agar Tube into the Bottom of each Agar Plate.

That's it. No Mad Hatter Tea Party with any of these Plates. He just poured

the Contents of each Tube of Top Agar over one Half of the appropriate

Bottom Agar Plate and allowed the Top Agar to solidify.

• "What are your Plaque Counts for each Plate of these Top Agar Slants?"

Hi normally the top agar should be 3ml instead of 1.5ml, and normally we would do Mad Tea Party to evenly distribute the top agar over the surface of the bottom agar. Would this have any effect on the result? The plaque count should be 30-300 so only 42 is valid.
